Abstract During tumor ontogeny and viral infection, heterologous subsets of CD8+ T lymphocytes provide immediate clearance durable protection. We utilized model antigen (HIV-1-Gag) expressing melanoma cell line parabiosis to investigate the role different components memory cells, including circulating cells (T cirm) tissue-resident RM), in lung metastasis first proved that RMgenerated by DNA vaccine intramuscular prime followed influenza-vectored intranasal boost immunization regime mediated potent long-lasting anti-lung immunity, whereas cirminduced vaccination combinations was not capable sufficient To further explain mechanism behind prime-boost immunization, we found elicited a large amount central CM) circulation preferentially differentiated into RMafter booster immunization. Single-cell RNA sequencing profile illustrated tissue microenvironment post mucosal reshaped differentiation CMelicited systemic enhanced anti-metastasis immunity. Thus, our research highlighted importance RMin protection provided insight RMestablishment vaccination.
